- Microbial cell factories (MCFs)
- MCF technology is a core aspect of green biomanufacturing. By leveraging the powerful and diverse biochemical reaction networks of microorganisms, microbial cells can be transformed into MCFs capable of producing various products from low-value renewable resources by remodeling and engineering metabolic pathways. We use genome-wide reorganization approaches, novel mutagenesis methods, and genomic library co-expression/co-expression to construct MCFs at the genome-wide level and use them for the production of saccharides.
- We develop cell factories by parsing the synthetic pathways of glycan biomolecules and reconfiguring them in microbial cells to enable the fermentative synthesis of target substances using renewable feedstocks. For example, in Yeast, we construct cell factories that utilize glycosyltransferases to synthesize carbohydrate antigens or natural glycans from scratch.
- Genetic Engineering
- The genetic engineering technologies we offer are centered around heterologous synthesis, yield enhancement, and custom synthesis. In the production of glyco-biomolecules, we provide CRISPR-based gene editing tools to genetically modify (transposon insertion and homologous recombination) the microorganisms that produce glyco-biomolecules. In this process, the CRISPR gene editing toolkit for Pseudomonas spp. can perform mutations, deletions, insertions, and single-base editing with nearly 100% efficiency. This method can greatly accelerate the screening of yield-enhancing genes and the construction of engineered bacteria.
- On the other hand, we employ abundant gene expression regulatory elements to aid in the production of glyco-biomolecules. In general, the production of glyco-biomolecules is closely related to the expression of enzymes; therefore, we regulate the expression of genes on a large scale by constructing enzyme expression promoter libraries.
- Cell Fermentation
- Cell fermentation is also an effective means of producing glyco-biomolecules. We scale up the production of glyco-biomolecules by analyzing microbial cells. For example, we analyze the utilization of different low-carbon compounds by yeast to construct yeast factories with a wider range of carbon sources. Next, we enhance the production and yield of yeast cells by mixing and scaling carbon sources.
- In addition, our research team has opened up the synthetic pathway for the synthesis of glyco-macromolecular compounds from glyco-small molecule compounds by introducing different synthetic pathways of glyco-macromolecules and regulating the endogenous glycogen synthesis and degradation pathway in yeast cells.

Technology: A bacterial model for polysaccharide production

Results: The authors constructed a model for the production of extracellular polysaccharides using Ramlibacter tataouinensis and successfully produced extracellular polysaccharides. First, the authors enriched the substance under small-scale conditions using lactate as an energy substance. Next, the culture medium of the bacteria was optimized to enhance the production of extracellular polysaccharides. Finally, the authors used trifluoroacetic acid to extract extracellular polysaccharides from the capsules and characterized them using chromatography.
