CD BioGlyco has extensive experience in developing Mammalian Chassis for Synthetic Biology (CHO cells, HEK293 Cells). CHO cells have been adapted for the Production of glycosaminoglycans, glycoproteins, and other substances. It is one of the ideal engineering Chassis. We provide a one-stop CHO glycoengineering service to help clients accelerate the production of glycoproteins and others, functional resolution, and other research. Based on our extensive experience in Chassis Development, we have a precise and scalable set of tools to characterize and control gene expression and function in CHO cells, culminating in stable CHO cell lines.
CHO cells dominate the production of glycoproteins, antibodies, etc. We design, construct, and characterize synthetic genetic pathways that ultimately enable CHO cells to produce a wide variety of glycoproteins, glycans, antibodies, and other assorted products predictably and stably.
Design: We combine computational system analysis and bioinformatics tools to rationalize the design of synthetic pathways.
Modification: We have various genome editing tools to precisely edit the CHO genome.
Validation of CHO mutants.
Site-specific recombination: This method relies on fungal or bacterial recombinases.
Clustered regularly interspaced short palindromic repeats (CRISPR) system: This method relies on DNA endonucleases, which are flexible enough to target DNA through short RNA rather than protein structural domains. It also mutates multiple alleles at the same time.
Transcription activator-like effector nucleases (TALEN) gene editing: This method is used to cut DNA at a specific location for gene editing.
Multiplexed genome engineering (MAGE): This method relies on oligonucleotide-mediated recombination and is a method for editing multiple genetic loci in parallel.

Results: In this study, a CHO cell was designed and constructed for the expression of antibodies initially produced in a murine cell line to produce murine glycans. Validated results showed that the two CHO cells constructed maintained stable production after 65 population doublings, while the glycosylation profile and function of the product matched the control. This study provides an effective strategy for engineering CHO glycans for glycan production.
